Abstract Approaches are needed for therapy of the severe acute respiratory syndrome from SARS-CoV-2 coronavirus (COVID-19). Interfering with interaction viral antigens angiotensin converting enzyme 2 (ACE-2) receptor is a promising strategy by blocking infection coronaviruses into human cells. We have implemented novel protein engineering technology to produce super-potent tetravalent form ACE2, coupled immunoglobulin γ1 Fc region, using self-assembling, tetramerization domain p53 protein. This high molecular weight Quad (ACE2-Fc-TD) retains binding spike and can complex plus anti-viral antibodies. The ACE2-Fc-TD acts as powerful decoy that out-performs soluble monomeric dimeric ACE2 proteins blocks both pseudovirus virus greatly enhanced efficacy. tetrameric promise be important development therapeutic against COVID-19.
